Bromantane: review, dopamine, doping and unknown risks
SupplementsImage de l’article

Bromantane, also called bromantan or Ladasten, is a synthetic molecule studied as a stimulant and treatment for asthenia, with no robustly established nootropic benefit in healthy adults. A Russian study reports 50–100 mg per day for 28 days in patients, but this studied dose is not a personal recommendation: no corresponding medicinal product was found in the French database, and bromantan is banned in competition by WADA.

Its marketing promises energy, endurance, calm and dopamine without tolerance or a crash. To compare this shaky evidence base with better-studied options, start with our overview of nootropics and their evidence.

The available literature does not support this level of certainty. The mechanisms involving tyrosine hydroxylase and dopamine come mainly from rats. One acute study included just ten healthy volunteers and primarily measured EEG and psychophysiological parameters. Russian studies on asthenia report improvements, but their assessment remains limited by access to the methods and their distance from optimising a healthy adult’s performance.

There is no robust clinical basis for defining a nootropic dose, sublingual route, cycle or absence of dependence. Bromantane is not an identified French medicinal product and is among the stimulants banned in competition by the World Anti-Doping Agency.

My assessment is unfavourable outside research or an authorised indication in a country where a clinician can prescribe it. I do not provide a consumption protocol because the human data do not allow a safe one to be developed. If your search concerns ADHD, start with so-called natural alternatives to Ritalin and their limitations.

What is bromantane?

It belongs to the family of adamantane derivatives. Ladasten is a medicinal product name cited in Russian studies, not a different molecule. Bromantan is notably the spelling used in the anti-doping list.

Where does the idea of an “actoprotector” come from?

The term actoprotector was used in Soviet pharmacology for substances purported to increase work capacity under strain without the activation typical of psychostimulants. This term is neither a European regulatory category nor a guarantee of mechanism.

Bromantane has been studied in contexts involving fatigue, performance and stress. Its adamantane structure does not make it clinically comparable to memantine or amantadine. Molecules sharing a chemical backbone can have different targets and risks.

The modern narrative adds a simple contrast: caffeine supposedly “borrows” energy, while bromantane supposedly builds new dopamine factories. This metaphor goes beyond the evidence. Caffeine primarily antagonises adenosine receptors. Bromantane modifies several pathways in animal models. Neither mechanism boils down to emptying or filling a tank.

Dopamine and tyrosine hydroxylase: what was actually measured

Tyrosine hydroxylase is an important enzyme in catecholamine synthesis. A rat study published in 2004 observed changes in tyrosine hydroxylase and DOPA decarboxylase gene expression in the striatum and hypothalamus after bromantane administration, along with variations in L-DOPA and dopamine.

Another rat study found regional changes in tyrosine hydroxylase messenger RNA and protein, dopamine content and hippocampal synaptic plasticity. Animal doses were high, and results depended on the brain region.

These studies do not establish the following claims:

  • correction of “low dopamine” in humans
  • increased receptor density
  • absence of tolerance
  • a lasting beneficial effect after stopping
  • repair after caffeine or short-form videos

Increasing an enzyme or neurotransmitter is not necessarily beneficial. Dopamine systems contribute to movement, learning and motivation, but also to impulsivity, insomnia and psychotic symptoms when disrupted.

The mechanism observed in rats supports a hypothesis. It cannot predict a user’s response, let alone improve it by adding tyrosine and vitamin B6.

What effects of bromantane have been demonstrated in humans?

The most directly relevant indexed human study included ten healthy volunteers after a single administration, compared with placebo. It observed EEG changes consistent with moderate alertness and reductions in certain tremor parameters. The main subjective measures and the performance of non-fatigued operators were not transformed.

Ten people are not enough to estimate a reliable average effect, rare toxicity, dependence or efficacy over several weeks. An acute study does not validate a course of use. Nor does it tell us whether blinding was maintained, whether a fatigued user responds differently or whether EEG changes translate into a useful improvement. Here, EEG is a marker, not proof of better brain health.

A 2009 placebo-controlled trial reports a reduction in neurasthenia symptoms over 28 days, followed by one week on placebo. Its abstract gives neither the dose nor the sample size: I will not reconstruct them from a seller’s website. The absence of a reported withdrawal syndrome over this short period does not demonstrate an absence of dependence after months.

A 2010 multicentre study describes 728 patients analysed across 28 Russian centres, with asthenic disorders and psychoautonomic syndrome. The abstract reports 50–100 mg per day for 28 days, followed by a follow-up one month after discontinuation. It describes improvement and 3% adverse effects, with no serious events reported, but does not describe a concurrent control group. These figures measure neither a benefit adjusted for the placebo effect nor the risk of a product used by a healthy adult.

Here are the three situations not to confuse:

Human dataOutcome studiedWhat cannot be inferred
Ten healthy volunteers, single administrationEEG, tremor and operator tasksLasting improvement in motivation or memory
Placebo-controlled trial, neurasthenia, 28 daysAsthenic symptomsEfficacy in every fatigued adult, absence of long-term dependence
728 patients analysed, multicentre studyClinical scores and short-term tolerabilityEfficacy versus placebo or general safety

Clinical asthenia is not a productivity plateau.

Bromantane has also been detected in sport and banned. An anti-doping ban establishes concern about performance enhancement or masking, not general therapeutic efficacy or safety.

Fatigue and motivation do not diagnose insufficient dopamine

Persistent fatigue can stem from insufficient sleep, sleep apnoea, anaemia, a thyroid problem, an infection, depression, a medication effect, low energy availability or overtraining. Loss of motivation may be contextual or psychiatric.

Feeling “drive” after taking a substance does not confirm its mechanism. Activation may mask the symptom while its cause persists. The user may then increase frequency, cut sleep short and add substances to counter anxiety.

Before looking for a stimulant, document the following:

  • duration and timing of fatigue
  • sleep quality and regularity
  • medications, alcohol, cannabis, nicotine and caffeine
  • mood, loss of pleasure and anxiety
  • diet and training load
  • signs such as breathlessness, palpitations, weight loss or fever

Unexplained fatigue lasting several weeks warrants a medical consultation. The article on the causes of persistent fatigue · FR helps you prepare useful information without concluding that dopamine is the cause.

For a comparison with another molecule often proposed for cognition, read the guide Noopept in France: evidence and risks. Citicoline and its human data · FR also allow comparisons around specific goals, without promising universal energy.

Tolerance, dependence and “no crash”

The absence of numerous reports does not prove the absence of dependence. A substance that sees limited use, is not monitored through French pharmacovigilance and is bought on the grey market generates incomplete data.

The available studies do not allow the following to be calculated:

  • frequency of insomnia, anxiety or palpitations
  • risk of tolerance after several weeks
  • symptoms after stopping
  • effects on mood in people with bipolar disorder
  • risk of psychosis with other stimulants
  • long-term liver and cardiovascular safety

Claiming that a de novo synthesis mechanism would prevent tolerance is a mistake. Homeostasis can act at multiple levels, including receptors, transporters, sleep and behaviour.

A “lasting effect” reported by a user may be a benefit, prolonged stimulation, sleep disruption or a mistaken attribution. Without a controlled trial, we cannot characterise it. The same caution applies to a “crash”: its absence the next day demonstrates neither recovery nor an absence of adaptation. Shortened sleep or delayed irritability may look like greater productivity before becoming a cost.

What dose of bromantane has been studied?

50–100 mg per day for 28 days appears in the abstract of the multicentre study in patients with asthenia. This historical exposure defines neither an effective dose for focus nor a safety threshold in healthy adults. The route is not specified in this abstract. I therefore do not turn this figure into an oral, sublingual or nasal regimen.

Administration routes, half-life and cycles: where does false precision begin?

Online guides often suggest oral and sublingual ranges, a fatty meal, an eleven-hour half-life and cycles of five days out of seven. These details create the appearance of established pharmacology.

Animal publications do not establish human sublingual bioavailability. I have not identified a clinical trial directly comparing oral and sublingual administration for nootropic use. Powder held under the tongue may expose you to an unpredictable dose and excipients not designed for this use.

There is also no trial showing that a 5/2 cycle prevents tolerance or protects receptors. Without a validated dose, recommending that you “start low” is not enough. A small amount of a poorly identified product remains an unknown exposure.

If you are considering countering stimulation with a serotonin precursor, first read about the risks and interactions of Griffonia and 5-HTP. A dopamine-versus-serotonin narrative does not validate a stack.

Stacks increase risk, not the quality of evidence

Combining bromantane, L-tyrosine and vitamin B6 rests on a mechanistic chain: more enzyme would supposedly require more substrate and cofactor. Yet the body regulates amino acid uptake, enzymes and neurotransmitters. More precursor does not guarantee a better signal.

The EFSA re-evaluation of vitamin B6 confirms the risk of neuropathy associated with excessive intake. Adding a cofactor is therefore not a risk-free precaution.

Combinations with tyrosine, methylene blue or psychoactive medications have no validated safety protocol for this use. Reasoning about dopamine cannot calculate their interactions. Have a doctor or pharmacist review all substances.

Combinations to avoid include the following categories:

  • methylphenidate, amphetamines and modafinil
  • MAOIs and methylene blue
  • antidepressants without medical approval
  • dopaminergic treatments
  • other research products

The absence of a published interaction is not permission. It often reflects a lack of studies.

Bromantane in France: authorisation, legality and doping

Checked on 13 October 2026: no occurrence of bromantane, bromantan or Ladasten was found in the official medicinal product and composition files of the French public medicines database, updated on 29 September. This database covers medicines marketed or having been marketed during the last three years in France. The absence of a result is therefore not an exhaustive inventory of all historical authorisations.

Above all, it does not constitute authorisation to sell or consume the substance as a supplement. “Not identified in pharmacies”, “not classified as a narcotic” and “legal in every situation” are three different claims. This article does not establish a general right to purchase, import or possess it.

A “chemical research” label is not a status permitting consumption. An HPLC certificate may relate to a different sample, be falsified or fail to test solvents, metals, isomers and stability. Buying exposes you to uncertain identity and concentration.

For athletes, the 2026 WADA list currently in force names bromantan under S6.A, a non-specified stimulant prohibited in competition. This category does not imply medical permission outside competition and provides no discontinuation interval guaranteeing a negative test.

In France, bromantan also appears in the Order of 18 March 2026 concerning substances whose possession may be penalised under the Sports Code. This text must be read alongside the conditions of the Sports Code, without automatically extending its penalties to every private individual.

If you have already taken bromantane

Do not add another product to “balance” dopamine, GABA or acetylcholine. Record the product, batch, time, stated amount and combinations. Avoid driving if you feel stimulated, confused or drowsy.

Call 15 or 112 for the following signs:

  • chest pain or feeling unwell
  • very fast or irregular heartbeat
  • extreme agitation or hallucinations
  • severe headache with neurological deficit
  • seizure
  • suicidal thoughts or dangerous behaviour

For persistent insomnia, anxiety or palpitations, seek medical advice promptly. Tell the healthcare professional about the lack of data on the substance and show them the packaging.

A measurable alternative to escalation

For a productivity plateau, first try two weeks without any new substance. Keep your wake-up time consistent, move your caffeine cutoff earlier, tackle one priority task before checking messages and measure the number of completed work blocks. If the result does not change, investigate sleep, mood and health rather than a “receptor that needs fixing”.

For acute performance, caffeine has human data and known pharmacology, which does not make it suitable for everyone. Its dose and timing must remain compatible with sleep. A beginner nootropic stack · FR must never be an excuse to pile on experimental products.

Le Biohacker’s verdict

Bromantane has interesting animal pharmacology, a small acute signal in humans and reported results in patients with asthenia. That is enough to explain the scientific interest, not to declare steady energy without a crash.

The benefits for asthenia are not established to a standard that would support a recommendation for a healthy adult. Psychiatric, cardiovascular, metabolic and dependence risks remain very poorly quantified even today. The grey market and anti-doping ban add tangible costs.

The best scientific, clinical and regulatory advice is therefore clearly not an arbitrarily more conservative dose. It is to avoid using bromantane outside research or an authorized medical setting, and to assess the cause of fatigue before stimulating its consequences.

Frequently Asked Questions (FAQ)

  • Is bromantane a stimulant without a crash?

We cannot claim that. A small acute study in ten volunteers observed EEG changes, but the available human data do not quantify efficacy, tolerability, dependence or withdrawal in healthy users.

  • Does bromantane increase tyrosine hydroxylase?

Rat studies show changes in tyrosine hydroxylase expression and dopamine synthesis. This proves neither dopamine restoration nor lasting benefits in humans.

  • Is bromantane authorised in France?

No bromantane medicinal product has been identified in the French public medicines database. The label research chemical is neither authorisation for consumption nor a guarantee of quality.

  • Is bromantane banned for athletes?

Yes, in competition. Bromantan is named in section S6, Stimulants, of the World Anti-Doping Agency's Prohibited List. Athletes subject to testing must avoid it.

  • What dose of bromantane has been studied?

A Russian multicentre study reports 50–100 mg per day for 28 days in patients with asthenia. Its abstract does not describe a concurrent control group. This figure is neither a personal recommendation nor a validated nootropic dose for healthy adults.

  • Bromantan, bromantane and Ladasten: what's the difference?

Bromantan and bromantane are spellings of the same substance. Ladasten is the medicinal product name used in Russian publications. None of these names guarantees the identity or quality of a product obtained outside medical channels.
